Soru 1950 working with circular paper chromatography was abel to detect arginine dihydrolas activity in hemolytic streptococci . sine aginine ,ornithin ,and citrulline differed in solubility in organic soven , each moved at a different rete on the paper ,thus showing distinctly separated spots when sprayed with ninhydrin.
Although the use of paper chromatography provided definitive results concerning the metabolism of arginine, the procedure was lengthy ,requiring several day . thin-layer chromatography [TLc ]is similer in principle to paper chromatography ; it provides moch more rapid results and achieves relaboratory we developed a TL c method for determining arginine dihydrolas activity in nonferrmentative organisms [13]. Using cellulose precoated sheets we were abel to detech arginine dihydrolas activity in these organisims in a few hours . we have also fund this rapid tlc method useful for speciating enterobacter[14].other invesgetors[15] have used tlc In detecting arginine dihydroles ativitty and have recommended modification of the sovent system to improve the medthod ,s sensitivity .
When performing arginine dihydrolase test using an arginine basel medium with an indicator it is important to (a) inoculate a control tube without arginine and(b) seal aech tube with paraffin or sterile mineral oil . the PH of the control tube should remain unchanged or shift to acid, depending on the medtod , and remain at that PH. Should the control tube become alkaline, the test must be repeated . in narrow tubes the mineral oil or paraffin seal may be omitted. However , at the top of the medium some alkalinity will be observed becase of oxidative metaboliam of peptones. A seal is not necessary when using definitive medtods such as TLC or other chromatographic procedures, since the arginine substrate and amino acid products are specifically indenified.
